I have been doing this for more than 3.5 years. Live in Windsor and work in metro DTW.
About the car with CAD licence plates, it is not only for residency, but Cdn customs will not allow you to drive a US plated vehicle if you live in Windsor (or anywhere in Canada). As a Cdn resident, you cannot drive a US plated vehicle. Exceptions to the above rule - you can obtain a "Commuter Conveyance Permit" if you will be driving your employer's car that may be US plated.
Sometimes, Cdn customs will let you through without a commuter permit if you have a credible letter from your employer stating you will be driving the company US plated car.
Also, the US customs will ask you more questions if you are a Cdn resident and drive a US car.
Crossing the tunnel or the bridge takes few seconds. It is not a big deal. To enter US, you need proper approved I797 (if on H1) and a valid I-94 along with a valid passport stamped visa (if your nationality requires it). Typically, there are more questions the first few times until the licence plate gets captured on the camera and receorded few times. My plate shows me as a frequent traveller and I barely get asked more than 1 question and that is "anything to declare?" !!
To enter Canada, you need the PR card. Again, the only question I get asked is how long I was away and the answer mostly is "since morning".
Once in a blue moon, the computer at the crossing will flag you for a random check (has happened to me 3 times in the last 3.5 years). They will tell you so and send you to secondary inspection where they go over your car, car documents and your documents and verify them all in theitrdatabase. This may happen both at the US and Cdn customs.

From Windsor downtown to DTW downtown, it is about 5 min.
From east city limits of Windsor to tunnel, it is about 20 min.
From tunnel to metro detroit area like Rochester Hills is about 45 min.
Most people refer to metropolitan area of detroit when they refer to Detroit rather than the city of Detroit itself.
Windsor and adjacent suburbs have a maximum 30 minute drive time to tunnel while from the tunnel, the drive time could extend to almost 1.5 hours to reach the limits of metro detroit. That kind of gives you an idea.
