Anyone who lives in Canada and wants to apply for Skilled worker immigration will have to send his/her application to Buffalo, USA.

As you have been admitted in Canada for a period of more than 1 year you can apply for immigration from Buffalo, USA. The only problem you will face is that if you are called for an interview in US. In this case it will be your responsibility to get a US visa.
It does not matter if your wife is in India as you are the principal applicant.
After your immigration process is complete all you have to do is go to the nearest border, cross over to US and return to Canada to complete the immigration process.
