It's between 8 hrs. - 10 hrs. depending on starting end end points.
Allow slightly longer time for border crossing because of long weekend.
Leave early because you'll meet the evening rush hour once you get into the NJ/NY area.

I just travelled last weekend....it was not even close to long weekend...took 3 hours to cross border...soooo much lineup....coming back made sense as was sunday & late evening...all shoppers from outlets were lining up but while going it didn't make sense....definitely keep 3 hrs on hand each way.
Are you staying in hotels or family????...if Family , don't drive to downtown, take commuter or whatever trains suit you & then hop in cabs to go to different places if you don't like walking too much, although its fun to walk, no matter how tiring.
If you want to enjoy downtown, best is to stay in hotel for atleast one night & walk around the financial district & grand central at night....its altogether a different beauty at night time.

If you take the toll route which is I-90, the return trip will cost you a total of $40 or a little less in 'tolls'.

On the highway at the Gas and food stops you will see some of the coupon books for hotels if you intend using them. Pick a couple of them up. If you have the Cell Phone, and if you have roaming, you can call the hotels and find out when you have to be there to get those rates. (Before what time)
If you have GPS system, then you can instruct it to avoid toll routes and it will give you the directions as you cruise the highways. So plan well because some of the routes are slow and congested and repairs might delay your arrivals.
If you are a Member of the CAA, get a TRIP TICK and a guide also.

Having done this run a few times, I can say that the toll route is the fastest (there's a reason it's a toll route)
I have tried alternative routes to work around the I-90 toll and just ended up wasting time and gas.
If you are in the mood for a scenic drive to avoid the toll, you can take highway 20 from Buffalo and then take 390 down towards NY/NJ.
The toll route will save you at least 45 mins.
