The Operational Bulletin 251 is only applicable to those who enters the port of entry from outside. I was already living/working in Canada on work permit and did my landing inside Canada. After working in Nova Scotia for abo two years, I have moved to Calgary and now living in Calgary for more than a year.
The guide system it seems is no more used by CIC. You may check the here

They do not issue a PNP entry into Canada, unless you found a suitable job and they approved your application based upon that selection that you made. You can always go back to the supervisor and make a request that they post you with suitable placement some where in that province, since you say you seem to like it. Mention all of these details and make it clear. Some of the jobs do get filled and see if you can find something equal or better. They must have a demand for your skills to approve your entry into Sasketchewan.

Since we all are trackable and can be foound here with in Canada, they know that you will be appearing at their door step some time in the future and they can leave a MARKER there and do a lot of damage to your life and career and so try avoiding such issues by approaching them in advance and getting a clearance from them first, prior to moving out of the province.
Alternately spend Three years there and complete your citizenship prior to your move.
