Can a TN visa holder register and operate a business in the US, while working on a full-time job and maintaining his TN status?

No. For every job you do, u will need a separate TN.

If a canadian citizen wants to start a business in the US, does he have to show any investment funds and business plan while applying for TN? Is there a separate TN (self-employed) category for that?

There is no "self employed" TN category. The job offer must be from an american employer. At the most what you could do is get a contract from an american company (and get a TN specific letter from them to get a TN) and then you can make them pay on the 1099 basis to your cdn corporation (incorporated in Canada).
I do not see any advantage in this approach though, unless you are already "incorporated" in Canada.
